Important Safety Information — Wegovy® Pill (oral semaglutide tablets)

The Wegovy® Pill contains semaglutide, the same active ingredient used in the injectable form of Wegovy®. The method of administration differs (oral tablet vs. injection), but the safety profile, boxed warning, and contraindications are based on semaglutide itself.
Boxed Warning: Risk of Thyroid C-Cell Tumors
In studies in rodents, semaglutide caused thyroid C-cell tumors, including medullary thyroid carcinoma (MTC). It is unknown whether semaglutide causes thyroid C-cell tumors, including MTC, in humans, because the human relevance of these rodent findings has not been determined.
Do not use the Wegovy® Pill if:
- You or any family member has ever had medullary thyroid carcinoma (MTC), or
- You have Multiple Endocrine Neoplasia syndrome type 2 (MEN 2).
Tell your provider right away if you notice any of these symptoms of a possible thyroid tumor:
- A lump or swelling in the neck
- Hoarseness that does not go away
- Trouble swallowing
- Shortness of breath
What the Wegovy® Pill Is Used For
The Wegovy® Pill is a prescription medicine used along with a reduced-calorie diet and increased physical activity in adults to:
- Reduce excess body weight and help maintain weight reduction long term in adults with obesity (BMI ≥ 30 kg/m²), or in adults with overweight (BMI ≥ 27 kg/m²) who also have at least one weight-related condition (such as high blood pressure, type 2 diabetes, or high cholesterol); and
- Reduce the risk of major cardiovascular events — cardiovascular death, non-fatal heart attack, or non-fatal stroke — in adults with established cardiovascular disease and either obesity or overweight.
It is not known whether the Wegovy® Pill is safe and effective in people under 18 years of age.
Who Should Not Use the Wegovy® Pill
Do not use the Wegovy® Pill if you:
- Have a personal or family history of medullary thyroid carcinoma (MTC);
- Have Multiple Endocrine Neoplasia syndrome type 2 (MEN 2); or
- Have had a serious allergic reaction to semaglutide or any of the ingredients in the Wegovy® Pill.
Do not use the Wegovy® Pill together with other semaglutide-containing products or any other GLP-1 receptor agonist medicine.
Before Taking the Wegovy® Pill, Tell Your Provider If You
- Have type 1 or type 2 diabetes;
- Have a history of pancreatitis;
- Have kidney problems;
- Have a history of diabetic retinopathy;
- Have severe problems with your stomach emptying (severe gastroparesis);
- Have ever had a serious allergic reaction (such as anaphylaxis or angioedema) to a GLP-1 medicine; or
- Are pregnant, planning to become pregnant, or breastfeeding.
Give your provider a complete and accurate medical history. Leaving out relevant information could lead to serious harm.
Pregnancy and Reproductive Planning
- The Wegovy® Pill should not be used during pregnancy. Weight loss offers no benefit during pregnancy and may harm an unborn baby.
- If you become pregnant while taking the Wegovy® Pill, stop taking it and tell your provider.
- Because semaglutide stays in the body for a long time, stop the Wegovy® Pill at least 2 months before a planned pregnancy.
- Breastfeeding is not recommended while taking the Wegovy® Pill.
A pregnancy registry monitors outcomes in women exposed to semaglutide during pregnancy. If you become pregnant while taking the Wegovy® Pill, you and your provider are encouraged to report the pregnancy to the Novo Nordisk pregnancy registry at 1-877-390-2760 or www.wegovypregnancyregistry.com.
Drug Interactions
The Wegovy® Pill slows stomach emptying, which can affect how other oral medications are absorbed. Use caution with oral medicines that have a narrow margin of safety or that require monitoring, and tell your provider about all the medicines you take, including:
- Insulin or insulin secretagogues (such as sulfonylureas), which can raise the risk of low blood sugar when combined with the Wegovy® Pill;
- Other GLP-1 receptor agonist or semaglutide-containing medicines;
- Other weight-loss products; and
- All prescription and over-the-counter medicines, vitamins, and supplements.
Serious Side Effects
Seek medical attention right away if you have symptoms of any of the following:
- Thyroid tumors — a lump or swelling in the neck, hoarseness, trouble swallowing, or shortness of breath.
- Inflammation of the pancreas (pancreatitis) — severe and persistent abdominal pain that may spread to the back, with or without nausea or vomiting.
- Gallbladder problems — upper-stomach pain, fever, yellowing of the skin or eyes (jaundice), or clay-colored stools.
- Low blood sugar (hypoglycemia) — especially if you also take insulin or a sulfonylurea. Know the signs (dizziness, shakiness, sweating, confusion).
- Kidney problems (acute kidney injury) — particularly if nausea, vomiting, or diarrhea leads to dehydration.
- Serious allergic reactions — swelling of the face, lips, tongue, or throat, or trouble breathing.
- Diabetic retinopathy complications — vision changes, particularly in people with type 2 diabetes.
- Increased heart rate.
- Risk during anesthesia or deep sedation — because the medicine slows stomach emptying, food may remain in the stomach. Tell your surgical or procedure team that you take the Wegovy® Pill ahead of any planned surgery or procedure.
Most Common Side Effects
- Nausea
- Diarrhea
- Vomiting
- Constipation
- Abdominal (stomach) pain
- Headache
- Fatigue
- Indigestion (dyspepsia)
- Dizziness
- Abdominal bloating
- Belching
- Gas (flatulence)
- Low blood sugar (hypoglycemia) in people with type 2 diabetes
- Stomach flu (gastroenteritis)
- Acid reflux (GERD)
- Common cold symptoms (nasopharyngitis)
Side effects are most common during dose escalation.
How the Wegovy® Pill Is Taken
- Take one tablet by mouth once daily, in the morning, on an empty stomach, with no more than 4 ounces of plain water. Do not take it with any other liquids.
- Swallow the tablet whole. Do not split, crush, chew, or dissolve it.
- After taking it, wait at least 30 minutes before eating food, drinking other beverages, or taking other oral medications.
- The starting dose is 1.5 mg once daily, increased gradually under your provider's guidance (typically 4 mg, then 9 mg, then a 25 mg maintenance dose), about every 30 days, to help reduce gastrointestinal side effects.
- Take only one tablet per day. If you miss a dose, skip it and take your next dose the following day.
- Do not change your dose without speaking to your provider.
Additional Safety Notes
- Do not combine the Wegovy® Pill with other GLP-1 receptor agonist medicines.
- Do not adjust your dose on your own.
- Follow your provider's titration and administration instructions carefully.
